چکیده

Enzyme-based Detoxification of Organophosphorus Neurotoxic Pesticides and Chemical Warfare Agents. (December 2007) Rory James Kern, B.S., Texas A&M University Chair of Advisory Committee: Dr. James Wild There are some 15,000 known organophosphorus chemicals. Some of these OP’s, including VX and paraoxon, demonstrate an acute neurotoxicity due to the inhibition of cholinergic enzymes. Organophosphorus chemical warfare agents and pesticide neurotoxins are subject to hydrolysis by OP degrading enzymes. To be useful as a bioremediation or anti-chemical warfare agent, the enzyme must be tailored for, and integrated into, a practical application platform. Several studies have established enzyme-based countermeasures, describing such diverse applications as decontaminating foams for surface remediation, encapsulating enzyme with liposome for in vivo therapy, enzyme attachments to surfaces for biosensors and development of a corn expression system for large-scale enzyme production. The goal of the research described here is to select, investigate and improve the operational potential of organophosphate-degrading enzymes including Organophosphorus Hydrolase (OPH, 3.1.8.1) and Organophosphorus Acid Anhydrolase (OPAA, 3.1.8.2). Using saturation kinetics, the catalytic efficiencies of these two major detoxification enzymes were characterized with substrates representing each class of OP neurotoxin, phosphotriester, phosphothioate and

Neurotoxic disorders of organophosphorus compounds and their managements.

Organophosphorus compounds have been used as pesticides and as chemical warfare nerve agents. The mechanism of toxicity of organophosphorus compounds is the inhibition of acetylcholinesterase, which results in accumulation of acetylcholine and the continued stimulation of acetylcholine receptors. Therefore, they are also called anticholinesterase agents. Organophosphorus pesticides have largely...

Compatibility of Cw Agent Degrading Enzymes with Disinfectants and Foams

Over the years our group developed a non-corrosive, non-toxic, environmentally safe and userfriendly chemical agent decontaminant (advanced catalytic enzyme system ACES) in a dry powder form. The ACES is capable of hydrolyzing/detoxifying organophosphorus (OP)-based neurotoxic chemical warfare (CW) agents, i.e. G-type, V-type, and related OP based hazardous industrial materials. Bacterial enzym...

Fluorescent Chemosensors for Toxic Organophosphorus Pesticides: A Review

Many organophosphorus (OP) based compounds are highly toxic and powerful inhibitors of cholinesterases that generate serious environmental and human health concerns. Organothiophosphates with a thiophosphoryl (P=S) functional group constitute a broad class of these widely used pesticides. They are related to the more reactive phosphoryl (P=O) organophosphates, which include very lethal nerve ag...

A fluorescence-based biosensor for the detection of organophosphate pesticides and chemical warfare agents

Recently elevated concerns of environmental contamination with organophosphate (OP) insecticides and the possible use of OP nerve agents by terrorists have spurred interest in developing more accurate and sensitive methods for field detection and discrimination of these neurotoxic agents. The analysis of OP neurotoxins typically requires sophisticated equipment, extensive sample preparation tha...

Bacterial detoxification of organophosphate nerve agents.

Bacterial enzymes have been isolated that catalyze the hydrolysis of organophosphate nerve agents with high-rate enhancements and broad substrate specificity. Mutant forms of these enzymes have been constructed through rational redesign of the active-site binding pockets and random mutagenesis to create protein variants that are optimized for the detoxification of agricultural insecticides and ...
